Movat Pentachrome Stain

Movat Pentachrome Stain Service: Analyze Multiple Tissue Components in One Slide

At iHisto, we offer expert Movat Pentachrome staining for precise visualization of elastic fibers, collagen, muscle, mucin, and fibrin — all in a single slide. Ideal for cardiovascular, pulmonary, and connective tissue research, our protocol ensures clean color separation with consistent results.

🧪 Overview

The Movat Pentachrome stain is a multicolor histological technique designed to distinguish five key tissue components simultaneously. This makes it invaluable for:

  • Evaluating blood vessels and heart valves

  • Mapping extracellular matrix (ECM) in fibrotic tissue

  • Assessing tissue composition in lung pathology, atherosclerosis, and tumor stroma

iHisto’s refined protocol ensures vivid and reproducible color separation in both human and preclinical tissue samples.

🎨 How the Movat Pentachrome Stain Works

Movat Pentachrome uses sequential staining steps to label:

  • Elastic fibers & nuclei → black

  • Collagen & reticulin fibers → yellow

  • Muscle → red

  • Mucin & ground substance → blue

  • Fibrin → bright red

This technique provides rich contrast and detailed spatial resolution, especially in vascular, cardiac, and pulmonary structures.

🧬 Step-by-Step Staining Process

  1. Fixation and Sectioning
FFPE tissue is sectioned at 4–6 µm thickness.

  2. Deparaffinization and Hydration
Slides are cleared through xylene and rehydrated into distilled water.

  3. Iron Hematoxylin Staining
Stains nuclei and elastic fibers black.

  4. Sequential Dye Application
Multiple acid dyes and Alcian Blue are applied to label fibrin, collagen, muscle, mucin, and other ECM elements.

  5. Dehydration and Coverslipping
Slides are cleared and sealed for long-term preservation.

⚠️ This is a complex stain — strict timing and reagent control are essential for consistency.

🔬 Applications of Movat Pentachrome Staining

Movat is ideal for comprehensive analysis of tissue remodeling, vessel structure, and matrix composition.

Common use cases include:

  • Cardiac pathology – infarct zones, myocardium, and arterial walls

  • Atherosclerosis research – fibrous cap and plaque morphology

  • Pulmonary fibrosis or emphysema – elastic fiber loss and matrix remodeling

  • Tumor stroma characterization – mapping mucin and collagen-rich zones

  • Tissue engineering – heart valve ECM integrity and recellularization analysis
